Biosci™ Lymphocyte Separation Tube for Human Peripheral Blood

The number of mononuclear cells (lymphocytes and monocytes) in peripheral blood is between 1.075-1.090 g/ml, which differs from that of red blood cells, multinuclear leukocytes, and platelets. The densities of red blood cells and granulocytes are relatively high (about 1.092 g/ml) and the density of platelets is between 1.030 and 1.035 g/ml.

Therefore, Human Lymphocyte Separation Medium is used to create a density gradient and the diluted whole blood is carefully layered on the separation medium. After centrifugation, the red blood cells and granulocytes settle at the bottom of the tube due to their high density. Since their densities are less than or equal to the density of the separation medium, lymphocytes and monocytes are found at the interface of the separation medium. There may also be a small amount of cells suspended within the medium. By aspirating the cells at the interface, mononuclear cells can be separated from peripheral blood.

This allows whole blood to be directly added to the tube, eliminating the need for pre-mixing with separation medium below the sieve.
